چکیده

Searching for homologous DNA molecules, pairing of the molecules and exchange of the strand with one of a counterpart duplex molecule are a central process for homologous recombination. RecA protein in Esherichia coli has been known to play a pivotal role in the processes. The recA gene was identified by A.J. Clark and his colleagues more than w x 30 years ago 1 and the first biochemical activities of the protein were described in 1978 by Roberts et w x w x al. 2 and Ogawa et al. 3 independently. Since then, the RecA protein has been the main focus in w x the field studying recombination 4,5 . The RecA Ž . protein binds to single-stranded DNA ssDNA in the presence of a high-energy nucleotide cofactor such as ATP and forms a typical right-handed helical

Extraction of Deep Phylogenetic Signal and Improved Resolution of Evolutionary Events within the recA/RAD51 Phylogeny

The recA/RAD51 gene family encodes a diverse set of recombinase proteins that effect homologous recombination, DNA-repair, and genome stability. The recA gene family is expressed in almost all species of Eubacteria, Archaea, and Eukaryotes, and even in some viruses. To date, efforts to resolve the deep evolutionary origins of this ancient protein family have been hindered, in part, by the high ...

The preference for GT-rich DNA by the yeast Rad51 protein defines a set of universal pairing sequences.

The Rad51 protein of Saccharomyces cerevisiae is a eukaryotic homolog of the RecA protein, the prototypic DNA strand-exchange protein of Escherichia coli. RAD51 gene function is required for efficient genetic recombination and for DNA double-strand break repair. Recently, we demonstrated that RecA protein has a preferential affinity for GT-rich DNA sequences-several of which exhibit enhanced Re...

RecA-like proteins are components of early meiotic nodules in lily.

Early meiotic nodules (also called recombination nodules) are proteinaceous structures about 100 nm in diameter that are associated with forming synaptonemal complexes (SCs) during early prophase I of meiosis. Although their function is unknown, early nodules may be involved in searching for DNA homology before synaptic initiation. Two potential components of early nodules are Rad51 and Dmc1 pr...

Regulation of bacterial RecA protein function.

The RecA protein is a recombinase functioning in recombinational DNA repair in bacteria. RecA is regulated at many levels. The expression of the recA gene is regulated within the SOS response. The activity of the RecA protein itself is autoregulated by its own C-terminus. RecA is also regulated by the action of other proteins. To date, these include the RecF, RecO, RecR, DinI, RecX, RdgC, PsiB,...

DNA pairing and strand exchange by the Escherichia coli RecA and yeast Rad51 proteins without ATP hydrolysis: on the importance of not getting stuck.

The bacterial RecA protein and the homologous Rad51 protein in eukaryotes both bind to single-stranded DNA (ssDNA), align it with a homologous duplex, and promote an extensive strand exchange between them. Both reactions have properties, including a tolerance of base analog substitutions that tend to eliminate major groove hydrogen bonding potential, that suggest a common molecular process unde...
